Munzy
Things I would check in order are... Fuses, voltage to central locking motors, motors themselves, mechanism for jams/sticking.
Bluebar
If this is an early model Focus with key barrel access to both front doors, try locking/unlocking from either side and observe if all the interior handles move when either door lock is turned to lock/unlock. This may indicate if a door latch fault is present and which one, ie one handle/latch not moving when the other latches move. Hope this makes sense? Cosd
Providing there are no blown fuses, sounds like it could be the actuator in the drivers door, this controls the locking to all the other doors. I doubt all your locking motors have packed up at the same time. It's been a long time but if I remember correctly the Fords operate a negative pulse locking system, so when you lock the door either by remote or key, the drivers door actuator should give a negative pulse to lock on one wire and a negative pulse to open on another wire (Sorry can't remember wire colours, possibly white with green stripe and white with blue stripe, but those colours are in the foot well and a little more difficult to find). Taking the door card off, if you have a 12v tester, but the clip end on a positive terminal (battery if that's all you can find) and prod all the wires on the actuator one by one whilst locking and unlocking; If it lights up on say locking, then that's good, if it lights up on another wire then again that means you have both sides working. Then it gets more complicated because you need to go through the loom to check for breaks, or to the other doors to see if they are receiving the signal. Someone may correct me here and say its a positive pulse, if thats the case, put your clip end of the tester on an earth and do the same checking for a positive pulse. Either way, you should get a negative or positive pulse coming out of the actuator. If you are getting nothing then it needs replacing.
